What is RT-PCR test for Covid-19 and how does it work?

Book a visit to a test site to have the test today. Test sites are open 7 days a week. Order a home test kit if you cannot get to a test site.

You need to get the test done in the first 8 days of having symptoms.

On days 1 to 7, you can get tested at a site or at home. If youre ordering a home test kit on day 7, do it by 3pm.

On day 8, you need to go to a test site – its too late to order a home test kit.

Whats In A Pcr Test Kit

Home test kits contain:

  • a vial containing a small amount of liquid this must remain in the tube
  • a clear zip-lock bag with an absorbent pad
  • a bag with a QR code
  • 3 stickers

Youll need to swab both your throat and nose. If youve had a nosebleed in the last 24 hours, swab the other nostril or wait 24 hours from when the nosebleed stopped.

If you have a nose piercing, swab the other nostril. If you have a piercing on both sides, remove the piercing on the side youre going to swab.

The fabric tip of the swab should not touch anything apart from the inside of your throat and nose.

Follow these steps if youre doing a PCR test at home.

  • Read the instructions that come with your test kit.
  • Before you begin, blow your nose.
  • Wash your hands with soap and water for 20 seconds or use hand sanitiser.
  • Place your test kit on a clean surface such as a table.
  • Remove the swab from the packet from the stick end. Do not touch the fabric tip of the swab or let it touch anything else.
  • Using a mirror, take a look at the back of your throat, at your tonsil area. Its important that you dont let the swab touch your teeth, tongue or gums.
  • Holding the stick end of the swab, open your mouth and rub the fabric tip of the swab over your tonsils for 10 seconds. Its normal to find this a bit uncomfortable. Remove the swab carefully without touching anything else inside of your mouth.
  • Clean your hands again.

    What Are The Uks New Travel Rules

    The government has now said that travellers heading into the UK will need to take a Covid test before their departure to try and limit the spread of the virus upon peoples arrival into the country.

    This can be either a lateral flow or PCR test.

    Travel from so-called red list countries where Covid rates are high require people to isolate for 10 days. Nigeria is the most recent addition to this list.

    Rules differ for fully vaccinated people and the non-vaccinated.

    Travel rules for the fully vaccinated

    When you have arrived in the UK, you must take a PCR test before your second day in the country and isolate until receiving a negative result.

    You also need to complete a passenger locator form, which must be shown as you check in to travel on any form of transport to the UK from abroad.

    Government guidance says: Youll need to show your form when you check in to travel or board your plane, train or ferry to the UK. You can submit the form any time in the 48 hours before you arrive in the UK.

    Travel rules for the non-vaccinated

    If you have not been vaccinated or are not fully vaccinated and you are travelling to the UK, then you will need to abide by the following rules:

    • take a Covid-19 test to be taken in the 2 days before you travel to England
    • book and pay for day 2 and day 8 Covid-19 PCR tests to be taken after arrival in England

    Matthew O’Toole has no intention of cancelling his one-day visit to Portugal – but he’s still not entirely sure of the rules, nor the likely disruption and extra costs.

    He’s collecting his young daughters to spend Christmas in the UK, with the plan being that the girls travel from Spain with their mother across the border to Faro airport.

    He told the BBC: “The girls live near the Portuguese border, so it’s easier for me to go from Gatwick and fly to Faro. I go to the airport, hang about, the girls get dropped off with me, we go back with them same day.

    “I haven’t had them here since Christmas 2019. I saw them for one week in August this year,” he said.

    This time he’s planning to collect them on 21 December and return them on 2 January. Mr O’Toole said that while a review of the rules is possible in the next couple of weeks, that is no help for his planning.

    “We’re up in the air on the tests. I have to test to go to Portugal and then we all have to test to get back into the UK. I have to do tests that I am fit to fly even though on the same day.

    “There’s also talk of a test to cross the border from Portugal to Spain. It’s going to cost a fortune, I just wish there was more guidance,” Mr O’Toole said.

Since coronaviruses dont have DNA, the first step of PCR testing is converting the viruss RNA into DNA in a process called reverse transcription. This is because DNA is a lot more stable than RNA. The PCR machine then makes millions of copies of the DNA by running multiple cycles . This process is called amplification and is extremely important in finding even the smallest amounts of DNA. As more cycles are run, more copies of the DNA are made doubling every time it is copied and making it easier to find. If the piece of DNA cannot be copied, there is no virus in the sample, or there is such a low amount that even this very sensitive test cannot detect it.

    Concerns have been raised over the new Omicron variant, which is beginning to spread more rapidly among people in the UK.

    The Government of Canada will notify travellers should it become mandatory to obtain COVID-19 molecular tests from specific accredited laboratories or facilities.

    What Are The Rt

    The quantification of mRNA using RT-PCR can be achieved as a one-step or a two-step reaction. The difference between the two approaches lies in the number of tubes used when performing the procedure.

    In one-step protocol, the reverse transcription and PCR components are mixed in a single tube and buffer at the same time.
